Neha Pendse began her acting career when she was a child. Her first movie was "Pyaar Koi Khel Nahin" in 1999. After that, she appeared in films like "Devdas." Pendse also started working on television. She made her debut in the show "Captain House," which was produced by Ekta Kapoor and Balaji Telefilms.

In 2016, she played the main character, Sanjana, in the comedy show "May I Come In Madam?" on Life OK channel, which ended in 2017. Pendse has also been a contestant on reality shows like "Comedy Dangal" and "Entertainment Ki Raat." In 2018, she hosted the comedy game show "Family Time With Kapil Sharma" alongside Kapil Sharma.

Pendse was also a participant in the twelfth season of the Indian reality TV show "Bigg Boss." However, she was evicted after four weeks on October 14 (Day 28). In 2021, she took over the role of Anita Vibhuti Narayan Mishra in the popular sitcom "Bhabiji Ghar Par Hain!" after Saumya Tandon left. Pendse left the show in January 2022.

She received much praise for her acting in the Marathi drama film "June" and won the Filmfare Award Marathi for Best Actress.
