Beyond modeling and features, Kalyani's movie career shines in South Indian cinema. She made her debut in the Telugu film Hello (2017) opposite Akhil Akkineni, earning the Filmfare Award for Best Female Debut – South. She has since excelled in hits like Varane Avashyamund (2020, Malayalam debut), Maanaadu (2021), Hridayam (2022), Thallumaala (2022), and the blockbuster Lokah Chapter 1: Chandra (2025), where she played a powerful superheroine/yakshi, making it one of the highest-grossing Malayalam films.

Ongoing projects include Genie (Tamil, with Ravi Mohan), Marshal, a new untitled Tamil women-centric film (Production 7), Lokah Chapter 2 (reprising Chandra, shoot from September 2026), and her anticipated Bollywood debut in the zombie thriller Pralay opposite Ranveer Singh (shooting 2026).
